General Information

These toxins are responsible for more than 90% of mushroom poisoning fatalities worldwide. At least nine types

of amatoxins have been identified, including α-amanitin, β-amanitin, γ-amanitin, ε-amanitin, amanullin,

amanullinic acid, and proamanullin. Among them, α-amanitin and β-amanitin are the primary toxins responsible

for severe toxicity and lethality.

This product is designed for the rapid detection of amatoxin residues in mushroom-related samples. The entire

testing process takes only 10 minutes, making it suitable for on-site rapid screening by various enterprises and

testing institutions.

Test Principle

This product adopts colloidal gold immunochromatography, the reaction zone is covered with quality control line

(C line) and detection line (T line), the T line is the binding of AMA and bovine serum albumin (BSA) (AMA-BSA),

and the binding pad is the binding of anti-AMA monoclonal antibody and colloidal gold (Ab-Gdd), when the

sample does not contain AMA, When Ab-Gdd extends to the T-line with liquid chromatography, it binds

specifically to AMA-BSA on the T-line to form a deep red line (T line), and excess Ab-Gdd continues to surge

upward to the C-line to form a shallow red line (C line). When the sample contains AMA, the AMA in the sample

competes with AMA-BSA on the T-line to bind Ab-Gdd, making the T-line light or no color and the C-line dark. To

determine whether the sample contains amatoxin.

Product performance

Sensitivity: β-amanitoxin Standard 4ppb; Alpha-amanitoxin standard 20ppb; γ-amanitoxin Standard 10 ppb.

Notices

1. The optimum operation temperature is 20-30℃.

2. The strip is a disposable product and cannot be reused.

3. Do not use expired test strip.

4. Do not touch the white membrane in the center of strip.

5. Do not reuse droppers provided in case the cross-contamination.

6. Do not mix the strips and diluent reagent with different lot numbers.

7.If test the standards, please request for the corresponding buffer. Tap water, distilled water, deionized

water should not be used as the negative control.

8.The solid impurity particles in the sample will lead to false positive results. Please discard the visible

particles in the samples. (If necessary, centrifuge the sample-diluent solution and collect the supernatant as

the testing solution).

9. Amatoxin is a high toxic substance, please operate with gloves.

10. Please contact the supplier if any problem in testing.

11. The test results are for reference only. For confirmation, please refer to the relevant national standard

methods.

Sample processing procedure

When sampling, ensure clean all utensils to ensure the accuracy of the test.

Fresh mushroom sample (5 times dilution) :

Take 1 g of the homogenized test sample and add it to 5 mL of the diluted solution in a 10 mL centrifuge tube.

Shake and mix well for 5 minutes. Let it stand or centrifuge for 1 minute. The supernatant is the test solution.

Dried mushroom sample( 20 times dilution) :

After grinding and homogenizing the test sample with a grinder, take 0.2 g and add it to 4 mL of the diluted solution

into a 10 mL centrifuge tube. Shake and mix well for 5 minutes. After thorough mixing, let it stand or centrifuge for 1

minute. The supernatant is the test solution. If the sample has a high water absorption rate, the diluent can be

appropriately increased.

Serum sample/mushroom soup sample (5 times dilution) :

Take 0.1 mL of serum or mushroom soup and mix it thoroughly with 0.4 mL of sample diluent. The resulting

mixture is the test solution.

If the mushroom soup sample is relatively turbid, it needs to be centrifuged at 5000 rpm for 5 minutes at room

temperature or passed through a 0.45 μm filter membrane for testing.

Vomit sample (5 times dilution) :

After centrifuging the vomitus at 5000 rpm for 5 minutes at room temperature, take the supernatant and adjust

the pH to 6.0-8.0 with 1M sodium hydroxide. Take 0.1 mL of the processed vomit and mix it evenly with 0.4 mL of

the sample diluent. This is the solution to be tested.

Detection procedure

When removing the test card, do not touch the reaction area or the sample area to ensure the accuracy of

the test.

Please read the instruction manual completely before testing, and recover the test strip and reagent to the

room (20 -30 ℃) before use.

2. Take 100 μL (4 drops from a pipette) of the solution to be tested and add it to the sample well of the test

card. Incubate for 10 minutes.

3. The results should be read at exactly 15 minutes, and interpretation at other times is invalid. Those who wish

to keep the results for a long time, please take photos for record.

Result Analysis

Negative result (-): The T line (test line) is darker than the C line (control line), indicating that the

concentration of amatoxin in the sample is below the detection limit or there is no amatoxin residue in the

sample.

Positive result (+): The color of the T line that is as dark as the C line, or lighter than the C line, or no color on

T line, indicating that the concentration of amatoxin in the sample is at or above the detection limit.

Invalid result: When the C line of the test strip is not colored, It may have been improperly handled or the

test strip has failed. Please check the instruction again, and retest with a new test strip.

Validity and Storage Conditions

Valid for 12 months. Avoid direct sunlight and store at 2 - 8 ℃.Do not freeze.

The lot number and expiry date are printed on the outside of the package.
